Protocol S.6
Silinization of Slides
Superfrost plus slides and standard slides can be silinized but I have found that superfrost slides work best so that protocol is outlined here.
Solutions
Reagents Needed
Superfrost Plus Slides
TESPA-3 aminopropyltrioxysilane (Sigma # A3648)
acetone
EtOH
Concentrated HCl
Procedure
Dip a rack of superfrost plus slides in 2N HCl (200 ml Q + 40 ml concentrated HCl) for 1 minute.
Rinse the slides for 1 minute in Milli Q water.
Rinse the slides in 100% Acetone for 1 minute.
Incubate in 100% Acetone containing TESPA for 1 minute.
Rinse briefly in 100% acetone.
Rinse briefly in 95% EtOH followed by 50% EtOH, 30% EtOH and several changes of Q.
Air dry at room temperature overnight and then store at 4° dessicated until use. They keep for several months.
